在Thrb B2亚型(ENSMUST00000022304)特定内含子1的下游，这个内含子增强子的ebox1和ebox2序列经历了改变，从CACCTG替换为GAATTC，从ACTGTACACAGGTGTTCA变为CAGTGCTAGCTTGCAGAC。最初，为了筛选，我们在第4内含子中插入了一个自我剪接的ACN载体(loxP位点+雄性特异性的ACE启动子驱动的Cre基因+诺卡因抗性基因座+loxP位点)。随后，这个载体在构建杂合体时自我删除，仅留下一个loxP位点位于增强子上游的边界(位置J:335506)。
