在exon 3的上游插入了一个loxP位点，并在neomycin抗性基因周围设计了自剪切锚定位点-lox2272，接着是反向排列的SA-2A-dTomato-polyA插件，随后在exon 3的外显子3下游，再插入了另一个loxP和lox2272位点。这个neomycin抗性基因通过自剪切被去除了。利用lox2272的Cre介导重组，反转了dTomato序列。(来源：J:329964)
