RMCE通过重组插入了cre-依赖的FLEX(翻转剔除)元件，其中包含了一个CAG启动子在FLEX元件之前，FLEX元件内是hM4Di-mKateT的编码序列，但方向与启动子相反。FLEX元件的下游是反向插入的兔球蛋白polyA序列，每个读框都有终止密码子。hM4D的ORF后面跟着6xHis标签和V5标签，但这些在固定组织的免疫组化中并未检测到。(来源：J:304133)
