The murine Fabp4 sequence was humanized by introducing point mutations to convert the 11 amino acids in exons 2 and 3 that differ between mouse and human FABP4 to human FABP4 sequences. A loxP site was inserted within intron 1 upstream of exon 2 and a FRT-neomycin-FRT-loxP cassette was inserted at the 3 end of exon 2. Resulted chimera were crossed to Flp recombinaseexpressing mice to excise neomycin selection cassette. (J:339978)
