使用CRISPR/Cas9技术，构建了一个包含AS-loxP/NeoSTOP/loxP小鼠Mecom基因cDNA，后面跟着IRES-EGFP-pA融合基因的靶向载体，这个载体插入了ROSA26基因座的内含子1。这个载体被注入到6NK7胚胎干细胞中。在内源性Gt(ROSA)26Sor启动子驱动的Cre重组酶作用下，Mecom和EGFP基因能够表达。携带这种等位基因的线系显示出较低的重组率。（来源：J:336140）
