这个靶向构建插入了IRES序列，后面跟着编码融合蛋白的mER:Cre:mER cDNA，该蛋白由两拷贝的鼠雌激素受体(mER)环绕Cre重组酶，以及一个由Frt重组酶座 flank 的Neomycin抗性(NeoR)选择标记，位于Fgf8基因的Eag1位点，距离Fgf8终止密码子36bp，以及内源性polyadenylation序列的5'端。经过电穿孔，线性化的靶向载体被导入小鼠胚胎干细胞并进行筛选。Flp重组酶剪除了NeoR标记。(来源：J:301046)
