The targeting vector is constructed based on the Ai9 vector from Addgene with (from 5' to 3') a CAG (CMV enhancer/chicken beta-actin) promoter, an FRT site, a loxP site, a mouse CD40 signal peptide, a myc tag, a G5 tag (five N-terminal glycine residues) fused to Thy1.1 cDNA, a polyA signal, a second loxP site, a FLAG tag, a Staphylococcus aureus transpeptidase sortase A (srta)-tag cDNA attached to a linker and a human PDGFRB transmembrane domain followed by a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE), a polyA signal, and an attB/attP-flanked PGK-FRT-Neo-polyA cassette.
