这个转基因是通过将神经肽U受体1（Nmur1基因）的第2和3个外显子替换为P2A-iCre-T2A-EGFP cassette，接着是FRT引导的Kanamycin cassette，然后在小鼠BAC RP23-403I3中实现的。FRT引导的Kanamycin cassette通过FLP介导的重组被去除。修改后的BAC构建被注入C57BL/6胚胎的卵母细胞核中，随机整合到小鼠基因组中。建立了Founder line 1，并后续保持（J:334817）。
