The targeting vector was designed with a CAG promoter, an FRT site, a loxP-flanked STOP cassette, an mCh sequence, a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE; to enhance the mRNA transcript stability), a poly-A signal, and an attB/att-flanked PGK-FRT-Neo-poly-A cassette was inserted into the Gt(ROSA)26Sor locus via electroporation of C57BL/6-derived embryonic stem (ES) cells.
