靶向载体设计用于替换CART编码外显子，通过插入来源于小鼠Cartpt的cDNA，连接到原始CART基因的起始密码子，后面跟着带有FRT-flanked诺卡因选择性 cassette的翻转酶（Flp）重组靶位。插入了一个含Tamoxifen诱导型Cre重组酶基因的载体，后面跟着驱动mCherry红色荧光报告基因表达的内源性核糖体进入位点（IRES）。杂交后代与携带Flp重组酶的生殖细胞系小鼠杂交，以去除诺卡因 cassette。在CART和Cre基因的起始密码子上游分别放置LoxP位点，以便通过Cre替换CART基因。随后，纯合子小鼠与仅在卵母细胞中表达Cre的小鼠杂交，实现Cre基因与内源性CART启动子的重组。（来源：J:333645）
