The targeting vector is designed to replace the endogenous STOP codon of the gene with a TimeSTAMP cassette with a STOP codon, and an FRT-flanked neomycin (neo) cassette. The TimeSTAMP (Time-Specific Tagging for the Age Measurement of Proteins) cassette contains, from 5' to 3';, N-terminal Venus fragment, a cis-cleaving hepatitis C virus (HCV) NS3 protease, C-terminal Venus fragment, and two hemagglutinin (2xHA) tags.
