这个靶向载体被设计成替换基因内的内源性终止子，用上一个SMASh套件，包含一个终止密码子，以及一个由loxP序列标记的诺如霉素抗性基因（neo cassette）。SMASh套件包含的是能切割的丙型肝炎病毒（HCV）NS3蛋白酶。通过Cre介导的重组，neo cassette被去除了。（来源：J:101977）
