靶向载体通过FLP剔除系统(FLEx)构建，包含从5'至3'的序列：lox2272、loxP座，一个包含Ank3最后一个外显子的FLEx插件(ENSMUSE00001264329位于ENSMUST00000182992.8)，一个FRT引导的neo cassette，pGHpA序列，以及反向排列的EGFP报告基因，后面跟着lox2272和loxP座，这些插入在内源性最后一个外显子下游。通过FLP介导的重组，neo cassette被剔除了。(来源：J:333091)
