2-4号外显子被替换为一个转录抑制序列(STOP)由loxP座 flank，前三个编码外显子(2-4)的cDNA序列，使用超稀有密码子，并包含p.G12D的致癌突变，接着是4号内含子的5'564bp，以及一个由FRT座 flank的抗氨苄青霉素基因 cassette。通过Flp介导的重组移除neo cassette。移除stop cassette是为了以稀有天然密码子为基础，实现突变型Kras的低水平表达。(来源：J:328899)
