在逐步定向过程中，插入了一个带有floxed neo cassette的序列在第8外显子下游。随后，去除了neo，留下一个loxP位点。接着，插入了FRT引导的neo cassette和另一个loxP位点在第5外显子上游。通过Flp介导的重组，移除了neo，使得5-8号外显子被标记为floxed。最后，通过cre介导的重组，删除了5-8号外显子。(来源：J:321165)
