在第3外显子(功能域)的位置替换了LacZ的序列，后面跟着一个终止密码和一个由Frt座点 flank 的Neo选择性载体。通过Flp介导的重组移除了Neo载体。修改后的基因座产生了由第1和2外显子以及LacZ融合的蛋白，但功能蛋白缺失。免疫印迹分析证实了在纯合突变小鼠的海马中，完全没有蛋白质表达。(来源：J:308995)
