TALEN-targeting using a homologous donor molecule created a G-to-A point mutation at chr7:142217981 (GRCm39) in the a multi-species conserved consensus sequence bound by a repressor, ZBED6, and created a silent A-to-G mutation at chr7:142217996 to disrupt one of the TALEN binding sites to prevent re-cleavage.
