在exon 1上游插入了loxP位点，通过同源重组在exon 2下游插入了loxP和FRT标记的neo cassette。通过Flp介导的重组，neo cassette被移除，exons 1和2被留下了门控区域。随后，Cre介导的重组去除掉了exons 1和2。（来源：J:242006）
