在exon 2的上游插入了一个loxp修饰的TK/neomycin选择座，然后在exon 2编码区域的非编码区内，再插入了一个第三个loxp位点。通过将cre重组酶转染到ES细胞中，去除了这个选择座，从而实现了exon 2的敲除。(来源：J:269358)
