这个转基因构建包含了来自一个携带人类JAK2基因的BAC的1-12号外显子，经过修改去除了N542和E543位点（AATGAA），这些来自一位真性红细胞增多症（PV）患者的插入。构建中包含一个反平行的lox66位点和一个lox71位点，分别位于12号外显子的前后，接着是JAK2基因13-25号部分cDNA以及polyA信号。通过限制酶酶切，lox位点之间的序列被倒转，形成一个可诱导的Flip-Flop转基因。在翻转状态下，无法产生完整的JAK2蛋白。（来源：J:236259）
