KOMP的PG00089_Y_3_F01靶向载体被电穿孔注入到ES细胞中。该载体包含FRT序列后面是lacZ序列，接着是loxP位点，第一个loxP后面是人β-actin启动子驱动的诺卡因抗性基因，SV40polyA，第二个FRT位点和第二个loxP位点。在103382899位置，第三个loxP位点插入在目标exons 12-16下游。因此，12-16关键exons被loxP位点夹在中间。Cre介导的重组会删除12-16exons和诺卡因抗性基因。(来源：J:314432)
