: The targeting vector contains a loxP-flanked neomycin inserted into intron 6 and an aspartic acid to histidine amino acid substitution at position 488 (D488H; GAT to CAT) inserted into exon 6. Cre-mediated recombination removed the neomycin cassette.The mutation corresponds to human tumor-associated missense mutations in the ankyrin domain that disrupts BARD1 recognition of H4K20me0 (histone H4 unmethylated at lysine 20).
