The CAG-a loxP site flanked neo-Westphal stop-FLAG-mouse Foxo1* cassette followed by an FRT site-framed IRESeGFP expression cassette were inserted into ROSA26 locus via homologous recombination in the ES cells. The Foxo1 cDNA has mutations of T24 and S316 to A and S253 to D which confers the constitutively nuclear localized Foxo1 transcripts. In presence of cre recombinase, a loxP site flanked neo-Westphal stop sequence is removed to allow expression of mutated Foxo1 gene driven by CAG promoter. (J:301891)
