在内皮1中插入了一个从5'到3'带有loxP位点的neo-stop cassette、一个rox位点的stop cassette，以及CAGGS启动子驱动的2A-NLS-ZsGreen（鸡β-actin启动子带有CMV增强子）。这些带有Gt(ROSA)26Sortm1(CAG-ZsGreen)Hamm标记的转基因小鼠与CAGGS Dre小鼠杂交，目的是去除rox位点的stop cassette。通过插入的间隔的floxed neo-STOP cassette，ZsGreen的表达仅在cre介导剪切去除了neo-STOP cassette时才得以激活。（来源：J:314807）
