通过同源重组，将一个含有从5'至3'方向的剪接接受器、floxed neo/stop cassette以及编码突变核定位的lacZ序列（序列870-1987位置有内含子重复，导致1993号位点插入TGA终止密码子，造成译码阶段偏移）插入到定位点。父母株Gt(ROSA)26Sortm1Etz与cre敲除载体杂交，以去除floxed neo/stop cassette。lacZ序列的内含子重复导致β-半乳糖苷酶分子部分失活，但在少数经历罕见随机同源重组的细胞中，会恢复功能正常的lacZ序列。来源：(J:278250)
