The targeting construct inserted the tdTomato openreading frame into the Calca locus between just 5 of the initiation codon in exon 2 and the noncoding exon 6. An frt-flanked Sv-Neo gene was also included for positive selection. After homologous recombination in ES cells, resulting chimeras were bred with FLPase mice to remove the Sv-Neo gene. (J:301067)
