构建的融合蛋白是rtTA*M2的tetracycline响应域(来自Clontech的pTet-on Advanced)和SDKid-1的KRAB-AB沉默域(Clontech的pTet-tTS),命名为rtTS(tetracycline-controlled trans-silencer)。SA70b的pROSA-CAGGS-attP50/B53-hygro-NLSLacZ载体来自ARTEMIS Pharmaceuticals。从SA70b中去除CAGGS启动子和内含子片段,并插入到修改过的pROSA26质粒中,形成了pROSA26CAG-PAS载体。rtTS的cDNA被克隆进pBigT,再转接到pROSA26CAG-PAS,最终得到了定向到Rosa26的CAG-LSL-rtTS载体(J:255811)。
