A bacterial artificial chromosome (BAC) containing the mouse Oprm1 gene (RP24-112H16) was modified so that the translational stop codon in exon 4 of the Oprm1 gene was replaced by a cassette comprising the T2A sequence, icre/ERT2 gene with stop codon and FRT-flanked neomycin-resistant gene (T2A-iCreERT2-FRT-Neo-FRT). The homologous recombination in the mouse ES cells generated a chimeric mouse which was crossed with CAG-FLPe mice to remove the FRT-flanked neomycin resistance cassette in the offspring. (J:302775)
