The transgene comprises the MHC class II (I-Ab)-restricted TCR specific for a peptide derived from the glycoprotein (G) of vesicular stomatitis virus (VSV), derived from T cell hybridoma 31.2.10.4 (leader sequence (L), the rearranged Va4, JaTA72, Ca). The TCR sequence was preceded by the H2-Kb promoter and followed by beta globin intron 2, poly(A) signal sequences and an Igh enhancer. This construct was co-injected with a construct containing a VSV-IND-G-specific, I-Ab-restricted, Vb2+ 31.2.10 T cell hybridoma-derived Tcrb gene (leader, Vb2, Db1.1, Jb2.1, Cb2) with the same upstream and downstream sequences.
