这个靶向构建包含了插入在Foxp3翻译终止密码子下游、内源polyadenylation信号上游的IRES-EGFP-iCre序列，以及位于9-10号外显子间带有FRT座点的PGK-neo cassette。在胚胎干细胞中，通过同源重组使用了这个构造。通过与Flp-deleter雌性小鼠杂交，去除了PGK-neo cassette。插入在9-10号外显子间的Frt元件导致了异常的3'剪接点，生成了Foxp3转录本的过早终止密码子。(来源：J:305614)
