The targeting vector replaced the D-Q52*01-J4 with the B1-8 promoter and the modified V-region of the M315 myeloma protein heavy chain (generated from the MOPC315 cell line) in which the Vlambda2 exon 2 was replaced with a synthetic Vlambda315 exon 2 containing the modification YST to FRN (residues 94-96; TACAGCACC to TTCAGAAAC) and an FRT-flanked neomycin resistance cassette. Flp-mediated recombination removed the selection cassette. (J:282821)
