这个靶向构建设计将P2A自切割肽序列和Cre/ERT2序列以框架方式连接到视网膜色素上皮65（Rpe65）基因的14号外显子3'端。Cre-ERT2融合基因包含一个连接了人雌激素受体配体结合域的Cre重组酶。非编码的15号外显子后面紧接着插入了一个FRT标记的诺卡因抗性（neo）基因座。这个构建通过电穿孔法导入了C57BL/6的FLP衍生的iTL BF1胚胎干细胞（ES）细胞。在ES克隆扩增过程中，靶向序列中的Neo基因座会被去除。（来源：J:310003）
