Site-directed mutagenesis was used to insert nucleotide substitutions in exon 11 of the locus, altering tyrosine to phenylalanine at codon 485 (Y485F-TAT to TTT) and a silent mutation, leucine to leucine at codon 487 (L487L -TTG to CTC) and introducing a Sac1 site. The substitutions were initially described as Y466F and L468L, respectively. In addition, a loxP and FRT-flanked neomycin-resistance gene was located downstream of exon 11. Flp-mediated recombination removed the FRT-flanked neo cassette. (J:303230)
