靶向载体被设计成在mRNA的5' UTR上游插入一个loxP位点，紧邻第一个编码外显子（exon 2）。随后在3' UTR下游插入第二个loxP位点，接着是一个由FRT引导的PGK-neo cassette，后面跟着第三个loxP位点。通过Flp介导的重组，FRT引导的neo cassette被移除，使得exon 2区域被标记为可剪接。(来源：J:287983)
