The BAC clone (bMQ458o12) contains the Glp1r gene flanked by 6 exons of the Dnahc8 gene at the 5' end and the first two exons of the Umod1 gene at the 3' end. A vector containing an optimized Kozak translation initiation sequence, a beta-globin poly-adenylation signal (BpA), the mApple fluorescent reporter, and an FRT-flanked kanamycin/neomycin resistance cassette is inserted into the start codon of the Glp1r gene within the BAC clone. The FRT-flanked cassettes were removed by transient transfection of a FLP recombinase-expressing plasmid. (J:292440)
