在反义方向，即4号外显子下游插入了一个报告基因 cassette，通过Cre介导的2号至4号外显子的删除，会使得tdTomato基因（报告基因）方向反转，从而被表达。为了提高报告基因 messenger的稳定性和获得更强的信号，加入了WPRE序列（木鼠肝病毒的后转录调控元件）。随后，选择 cassette 通过FlpCre突变株进行了去除。（来源：J:94077）
