靶向载体被设计成插入一个HaloTag基因组序列（如下）、TEV酶识别序列、一个在第一个编码外显子（exon 1）上游的in-frame loxP-STOP-IRES-FLAG-loxP座，以及基因1的内含子1中的FRT引导的PGK-neo座。HaloTag是一个由罗氏罗德罗氏菌(DhaA基因)编码的33千道尔顿的卤代烷脱卤酶，它经过基因改造，能与合成的卤代烷配体形成不可逆共价键。通过Flp介导的重组，FRT引导的neo座被移除了。（来源：J:283893）
