在第27号内含子中插入了loxP位点和FRT位点，伴随着一个诺卡因抗性基因座。接着在第28号内含子中，插入了另一个loxP位点和一段带有E1605Q变异（替换为谷氨酰胺密码子的谷氨酸）的exon 28拷贝。通过后续的flp介导重组，去除了诺卡因抗性基因座，形成一个条件性可操作的等位基因，其中包含野生型的exon 28（最后一个编码外显子）被敲除。当cre介导的重组发生时，删除了含野生型的敲除片段，而表达的是突变的exon 28。来源：J:279595
