将loxP序列和Neo cassette，前后带有两个Frt座标，插入在第12外显子上游331bp的位置。接着，将小鼠Hprt基因的内源性核糖体进入位点（IRES）和剪接起始点（SD）序列插入Neo cassette，用于poly-A捕获策略。另一个loxP座标则被引入在第12外显子下游185bp的位置，目的是在Cre介导重组后消除潜在的跨膜域。（来源：J:279639）
