这个靶向载体设计如下：从5'至3'有CMV-IE增强子/鸡β-actin/兔β-globin的融合启动子(CAG)，一个FRT位点，一个含loxP切割位点的停泊子，dCas9序列(dCas9-NLS-p300，下文详述)，一个1xFLAG标签，WPRE序列，bGHpolyA信号，以及attB-PGK-frt-Neo-polyA-attP插入片段。整个构建被插入Gt(ROSA)26Sor基因座的1号和2号外显子之间。这里的dCas9由优化过的、无内切酶活性的Streptococcus pyogenes Cas9蛋白DNA与人E1A关联蛋白p300的HAT核心域（EP300）的核定位信号和N端部分连接而成。(来源：J:342286)
