第五号染色体上，起始于136,150,952位置的外显子(在Orai2-001转录本中是第三个外显子，在Orai2-003转录本中是第五个外显子)被LoxP序列包裹。插入了一个FRT引导的YFP报告基因(En2剪接点、2A自切割蛋白、YFP编码序列)和抗氨苄青霉素抗性基因的STOP-Neo cassette，位于可剪切区域上游。通过Flp介导的重组移除了YFP报告基因和Neo cassette，随后Cre介导的重组删除了第三外显子和第五外显子。qPCR证实了腹膜巨噬细胞中转录本的缺失。(来源：J:278536)
