Cas9 mRNA和单链向导RNA(sgRNA)，针对小鼠Mettl4第6外显子的目标，被注入到囊胚期胚胎的细胞质中，以诱导删除。注射后的胚胎被植入CD-1代孕小鼠中，然后对幼崽进行筛选以检测删除情况。一轮注射后，五只小鼠出生，其中四只带有第6外显子的缺失。选择了三只作为实验分析的创始个体。这个等位基因有第6外显子的缺失，导致了翻译框架的移位，从而丧失了催化区域，预计产生一个甲基转移酶的敲除(KO)等位基因。(来源：J:278437)
