在7号染色体GRCm38的位置，127,841,278上游插入了loxP位点，接着在内含子4中插入了另一个loxP位点，后面跟着由磷葡萄糖激酶启动子驱动的诺卡因抗性基因（PGK-Neo），它被两个FRT序列 flank。通过Flp介导的重组，去除了诺卡因选择 cassette。这样，exons 1-4区域被标记为可删除。随后在生殖细胞中Cre表达导致loxP标记的基因组序列被消除，产生了敲除等位基因。来源：J:275367
