设计的DAT-ires-Flpo靶向元件包含一个loxN介导的诺卡因抗性(neo)基因座，随后是内质网进入信号(IRES)和Flpo重组酶序列，位于转运体家族6(神经递质转运器，多巴胺)成员3(Slc6a3)基因的终止密码子之后。正确靶向的ES细胞被注入囊胚，产生的杂合雄性与CMV-cre小鼠杂交以剔除neo基因座，参考文献为(J:101977, J:340041)。
