Zinc finger nuclease (ZFN)-mediated genome editing was used in NOD.Idd9.3 zygotes to target the second coding exon of the Tnfrsf9 gene (target sequence CCCTCCAAGTACCTTctccaGCATAGGTGGACAGCCGAA). Progeny were sequenced at the locus targeted by the ZFN to map the insertion. Founder line 24 contains a two base-pair insertion (aa) at the target site, resulting in a premature stop codon.
