在Dbh基因座的起始密码子后，通过同源重组插入了rox-flanked转录终止子、cre cDNA、兔β-球蛋白poly(A)序列以及attB/attP介导的诺卡因抗性 cassette。构建的重组克隆短暂转染了pPGKPhiC31obpa，以在移植前去除诺卡因抗性元件。然后将这些细胞注入囊胚以产生转基因小鼠。经过生殖传递，杂合体与表达Dre的个体杂交，目的是去除rox-flanked的终止子片段。（来源：J:285301）
