采用双靶向策略，目标去除包含6个酒精脱氢酶基因的250kb区域。构建了一种载体，将LoxP位点和抗羟胺霉素基因插入Adh7基因的第3内含子，另一种载体则设计用于Adh5基因的第4内含子插入Neomycin抗性基因和LoxP位点。这6个Adh基因位于3号染色体，顺序为Adh7、Adh1、Adh6a、Adh6b、Adh4和Adh5，该区域没有其他基因。Cre介导的重组去除了含LoxP的区域，以及Neomycin和Hygromycin的 cassette。(来源：J:271228)
