所有三种主要亚型(A、B、C)的前两个外显子都由loxP位点包裹，上游插入了一个含loxP的诺姆西林筛选载体，诺姆西林筛选载体的N端有一个loxP。通过Cre介导的重组，这个诺姆西林载体被剔除，产生了条件性等位基因。去除两个含floxed外显子会消除所有三种亚型共有的生长抑制性Nogo-66氨基酸链的表达，预测对C亚型为完全缺失，A和B亚型为严重减弱。(来源：J:268491)
