在ATG起始点大约200bp上游插入了loxP位点，通过同源重组在exon1下游插入了带有FRT标记的neo cassette和另一个loxP位点。通过重组酶介导的重组，exon1和neo cassette都被移除了。免疫组织化学分析证实了纯合小鼠大脑中没有蛋白表达(来源：J:266702)。
