A loxP site and an FRT site flanked neomycin resistance gene cassette (in the same transcriptional direction as the targeted gene) were inserted into intron 1. A second loxP site was inserted into intron 1 of upstream neighbor Peg3 (with which it shares a bidirectional promoter), in the opposite orientation of the first loxP site, upstream of the Peg3-DMR (differentially methylated region).
